Linkage analysis was performed on a population of (CBA/N x DBA/2)F1 x CBA/N backcross animals to map QTLs associated with susceptibility to cerebral malaria. Parental strain CBA/N is susceptible to infection with Plasmodium berghei ANKA and die within 14days post-infection. Parental strain DBA/2 and (CBA/N x DBA/2)F1 hybrids are relatively resistant to P. berghei ANKA infection. 65 polymorphic markers covering 70% of the genome were used in the genome scan. Male backcross animals were infected with 1 x 10(6) P. berghei ANKA infected erythrocytes at 8-10 weeks of age.

A locus named Cmcs (cerebral malaria susceptibility in CBA/N) mapped to mouse Chromosome 17 near the H2 locus. Peak linkage occurs at 18.44 cM near D17Mit28 (P<0.0001). CBA/N-derived alleles confer susceptibility to P. berghei ANKA infection at Cmcs whereas DBA/2-derived alleles confer resistance. Candidate genes mapping near Cmcs include Tnf (19 cM), Lta (19 cM), H2-K, H2-Aa (18.65 cM), and H2-Ea (18.7 cM).
