Mapping and Phenotype information for this QTL, its variants and associated markers

J:93737

Linkage analysis was performed on 222 female animals from a (NZB/BlNJ x BALB/cByJ)F2 intercross to identify QTLs associated with susceptibility to autoimmune hemolytic anemia (AHA) and systemic lupus erythematosus (SLE). 96 polymorphic markers at an average spacing of 17 cM were used in the genome scan. Parental strain NZB/BlNJ is susceptible to autoimmune hemolytic anemia and exhibits elevated serum levels of anti-red blood cell antibodies compared to BALB/cByJ. NZB/BlNJ is also susceptible to systemic lupus erythematosus and exhibits elevated serum levels of antinuclear antibodies compared to parental strain BALB/cByJ.

Linkage to anti-red blood cell IgM antibodies at 6 months of age mapped to 4.1 cM - 5.4 cM on mouse Chromosome 7 (P=1.1 x 10-3). This locus is named Arigm2 (anti-RBC IgM antibodies 2). Linkage to anti-red blood cell IgM antibodies at 8 months of age mapped to 6.8 cM - 8.2 cM and is named Arigm3 (P=5.0 x 10-4), and anti-red blood cell IgM antibodies at 10 months of age mapped to 58.3 cM - 59.7 cM and is named Arigm4 (P=3.0 x 10-3). Previously identified autoimmune loci Sle3 (15 cM), Lbw5 (23 cM), and Yaail (9 cM) map near Arigm2 and Arigm3. Aem2 (27 cM) also maps near Arigm2, Arigm3, and Arigm4. Linkage anti-red blood cell IgG antibody production at 8 months of agemapped to 13 cM - 13.6 cM (P=4.7 x 10-3). This locus is named Arigg7 (anti-RBC IgG antibodies 7). Previously identified autoimmunity loci Aem2 (27 cM), Sle3 (15 cM), and Yaail (9 cM) map near Arigg7. NZB/BlNJ-derived alleles confer increased anti-red blood cell antibodies at all chromosome 7 loci.
