Linkage analysis was performed on 16 LGXSM (LG=LG/J; SM=SM/J) recombinant inbred (RI) lines to identify QTLs associated with obesity and diabetes traits. Parental strain LG/J exhibits a larger body size compared to parental strain SM/J and these two strains also differ in obesity and diabetes related traits in response to dietary fat. 506 microsatellite markers at an average spacing of 2 to 3 cM were used in the genome scan. 39 loci were mapped but only those reaching genomewide significance are reportedhere.

On mouse Chromosome 1, an interval between D1Mit43 (73 cM) and D1Mit103 (76 cM) shows linkage to total fat pad weight, reproductive, renal, and mesenteric fat pad weights, body weight at necropsy, tail length, and kidney weight. This locus is designated Obsty1 (obesity 1). LG/J-derived alleles confer increased fat pad weights and increased values for the other traits. Obsty1 also shows suggestive linkage to inguinal fat pad weight and leptin levels. A second interval from D1Mit196 (71.5 cM) to D1Mit507 (87.8 cM) is linked to cholesterol levels and also shows suggestive linkage to heart weight. LG/J-derived alleles confer increased cholesterol at this locus. This interval is included in the Obsty1 locus but could possibly be a separate locus. Potential candidate genes mapping near Obsty1 are Avpr1b, Insig2, Hdlbp (55.3 cM), Capn10, Pfkfb2, Cd55 (67.6 cM), Dbi, and Tnfrsf11a.
