Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 16 LGXSM (LG=LG/J; SM=SM/J) recombinant inbred (RI) lines to identify QTLs associated with obesity and diabetes traits. Parental strain LG/J exhibits a larger body size compared to parental strain SM/J and these two strains also differ in obesity and diabetes related traits in response to dietary fat. 506 microsatellite markers at an average spacing of 2 to 3 cM were used in the genome scan. 39 loci were mapped but only those reaching genomewide significance are reported here.

On mouse Chromosome X, an interval between DXMit55 (1.4 cM) and DXMit10 (63 cM) shows linkage to glucose tolerance at 20 weeks of age. This locus is named Dbts2 (diabetes 2). SM/J-derived alleles confer increased glucose tolerance at this locus. Dbts2 also shows suggestive linkage to fasting glucose at 20 weeks. Potential candidate genes mapping near Dbts2 are Gpr50 (26 cM), Avpr2 (29.5 cM), G6pdx (30 cM), Pgr15l, Tsc22d3, Irs4 (58 cM), Capn6 (58 cM), and Mbtps2.

A second interval between DXMit121(67 cM) and DXMit186 (69 cM) shows linkage to total fat pad weight, mesenteric fat pad weight, leptin and cholesterol levels in female animals on a high fat diet. This locus is named Obsty5 (obesity 5). SM/J-derived alleles confer increased fat pad weights and increased leptin and cholesterol at Obsty5. Obsty5 also shows suggestive linkage to renal and reproductive fat pad weights in female animals on a high fat diet, body weight at necrosis, insulin levels, and spleen weight. Obsty5 also shows suggestive linkage to liver weight on a high fat diet. Potential candidate genes mapping near Obsty5 are Grpr (70 cM), Ace2 (70.5 cM), Tlr8, and Tlr7.
