Mutation details: The transgene contains the V-J exon from the rearranged Tcra gene of the (C3H/HeJ x DBA/2J)F1-derived, IA-d-restricted, rabbit immunoglobulin-specific CDC35 T-lymphocyte cell line. This V-J exon, in which Tcra-V8 is joined to Tcra-J37, was inserted into the 2B4alphaEH vector between a 2-kb germline DNA fragment from the Tcra-V11 5' flanking region, which contains the natural Tcra-V promoter, and a chimeric sequence comprising: a 681-bp fragment containing the immunoglobulin heavy chain enhancer; approximately 100 bp from the 5' end of the Tcra-J-C intron; and a 9.8-kb germline DNA fragment including 6 kb of Tcra-C 5'-flanking DNA, all of the Tcra constant-region exons, and 0.5 kb of 3'-flanking DNA.
