Linkage analysis was performed on a population of (OcB-9/Dem x O20/A)F2 animals to identify QTLs associated with T lymphocyte proliferation in response to cytokines IL-2 and IL-4. Parental strain OcB-9/Dem is a recombinant congenic strain carrying B10.O20/A donor DNA segments on an O20/A genetic background. OcB-9/Dem differs from O20/A at DNA segments on 10 different chromosomes. 26 microsatellite markers spanning these differential regions were typed in the F2 population. Recombinant congenic strain OcB-9/Dem exhibits a stronger T lymphocyte proliferative response to IL-2 and IL-4 compared to background strain O20/A.

A locus on mouse Chromosome 4 near D4Mit126 (71 cM) shows linkage to lymphocyte response to hIL-2, low levels of mIL-2, and high levels of IL-4. This locus is named Cinda3 (cytokine induced activation 3). The QTL interval spans D4Mit54 - D4Mit126. Heterozygous animals show the highest T lymphocyte proliferative response and animals homozygous for O20/A-derived alleles show the lowest proliferative response. In addition, female animals exhibit a higher response to cytokines compared to males. A previously identified lung tumor QTL, Sluc6 at 67 cM, maps to the Cinda3 region. Ssic1 (67 cM) and Pla2g2a (68 cM) also map near Cinda3. Tlsr3 (63 cM) andStmn1 (65.7 cM) map more centromerically to Cinda3.

A locus on mouse Chromosome 19 near D19Mit9 (48 cM) shows linkage to T lymphocyte proliferation induced by low levels of IL-4. This locus is named Cinda4 (cytokine induced activation 4). Animals homozygous for B10.O20/A-derived alleles exhibit the highest lymphocyte response. A previously identified lung tumor QTL, Sluc1 (47 cM), maps to the Cinda4 region. Mxi1 (49.5 cM) and Csf2ra (51 cM) also map near Cinda4.

A locus on mouse Chromosome 8 nearD8Mit3 (10 cM) and D8Mit65 (22.5 cM) shows linkage to T lymphocyte proliferation induced by high levels of IL-4. This locus is named Cinda5 (cytokine induced activation 5). The B10.O20/A-derived allele confers dominantly inherited increase in T lymphocyteproliferative response to IL-4. Female animals exhibit a stronger T lymphocyte response to IL-4 compared to males. A previously identified lung tumor QTL named Sluc20 (20 cM) maps near Cinda5. No potential candidate genes have been identified for Cinda5.
