Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 100 female animals from a (NZW/LacJ x BALB/cJ)F1 x NZW/LacJ backcross to identify QTLs associated with autoimmune traits. 77 polymorphic markers were used in the genome scan.

Linkage to autoantibody production mapped to 52 cM on mouse Chromosome 1 near D1Mit48 (P<0.001). This locus is named Baa2 (BALB/c autoimmunity 2). BALB/cJ-derived alleles at Baa2 confer increased IgM anti-ssDNA and anti-histone antibody production. Suggestive linkage to IgG anti-ssDNA antibody production mapped to 103 cM on mouse Chromosome 1 near Tgfbm2 (formerly D1Mit17). Previously identified autoimmunity QTLs mapping near this locus are Sle1 (88 cM) and Lbw7 (90 cM).

Linkage to autoantibody production mapped to 27 cM on mouse Chromosome 9 near D9Mit22 (P<0.001). This locus is named Baa1 (BALB/c autoimmunity 1). BALB/cJ-derived alleles at Baa1 confer increased IgM anti-ssDNA and anti-histone antibody production.

Linkage to IgG anti-ssDNA antibody production mapped to 28 cM on mouse Chromosome 11 near D11Mit207 (P=0.0031). Animals homozygous for NZW/LacJ-derived alleles at D11Mit207 are more likely to exhibit elevated serum IgG anti-ssDNA antibodies. A previously identified autoimmunity QTL mapping near this locus is Lbw8 (29 cM).

(NZB/BlScr x NZW/LacScr)F2 intercross, which differs from the mapping population used here, we consider the current study a separate experiment and have named the QTL identified here Nwa3, New Zealand White autoimmunity 3.

The strongest linkage to IgG anti-ssDNA antibody production mapped to the H2 locus on mouse Chromosome 17 (P=0.0004). Animals heterozygous at the H2 locus exhibit increased susceptibility to elevated IgG antibodies.

Linkage to IgG anti-ssDNA antibody production mapped to 38 cM on mouse Chromosome 19 near D19Mit11 (P=0.0005). This locus is named Nwa2 (New Zealand White autoimmunity 2). Animals homozygous for NZW/LacJ-derived alleles at Nwa2 are more likely to exhibit elevated serum IgG anti-ssDNA antibodies. Nwa2 also shows weak linkage to IgG anti-histone antibodies (LOD=1.09).

Suggestive linkage to IgG anti-ssDNA antibody production mapped to 32 cM on mouse Chromosome 16 near D16Mit5. Animals homozygous for NZW/LacJ-derived alleles at these loci are more likelyto exhibit elevated serum IgG anti-ssDNA antibodies. This locus is tentatively named Nwa1 (New Zealand White autoimmunity 1).

Suggestive linkage to IgG anti-histone production mapped to 20 cM on mouse Chromosome 14 near the Tcra locus (P=0.0017). Animals homozygous for NZW/LacJ-derived alleles at this locus exhibit increased IgG anti-histone production.
