Mapping and Phenotype information for this QTL, its variants and associated markersJ:95337Dice1, a previously identified QTL at 8 cM on mouse Chromosome 16 controlling IL-4 production and CD4+ T helper cell (Th2) commitment was resolved into 2 separate QTLs named Dice1a and Dice1b using interval specific congenic strains derived from a B10.D2-H2d/oSnJ donor strain and a BALB/cAnN background strain. Background strain BALB/cAnN exhibits high Th2 bias and susceptibility to infection with the parasite Leismania major compared to donor strain B10.D2-H2d/oSnJ. A panel of 9 congenic strains wereconstructed that formed an overlapping tiling path of B10.D2-H2d/oSnJ-derived donor DNA segments with 92% coverage of chromosome 16. Dice1a maps within the 95% confidence interval of Dice1. The minimum QTL interval of Dice1a spans from D16Mit142 (4.3 cM) to D16Mit164 (8 cM). Dice1a shows linkage to IL-4 levels and Th2 cell bias. The Dice1a interval is 3.2 Mb in size and contains 35 genes. Potential candidate genes are Carhsp1 (1.7 cM), C2ta, and Socs1 (3.4 cM).Dice1b maps distal to Dice1a. The minimum QTL interval ofDice1b spans from D16Mit138 (31 cM) to D16Mit27 (45.5 cM). Dice1b shows linkage to IL-4 levels and Th2 cell bias as well as resistance to infection with Leishmania major. Resistance to L. major maps within the Dice1b interval but may or may not be controlled by a separate gene. The Dice1b interval spans 27.6 Mb and contains 145 genes. Potential candidate genes are Bbx, Cblb, and Pit1 (42.5 cM). A previously identified cytokine production QTL named Cypr1 (30 cM) maps near Dice1b. |
