Linkage analysis was performed on 184 animals from a (BALB/cOlaHsd x CBA/CaOlaHsd)F2 intercross to identify QTLs associated with susceptibility to infection with the bacteria Streptococcous pneumoniae. Parental strain BALB/cOlaHsd is resistant to S. pneumoniae infection whereas parental strain CBA/CaOlaHsd is susceptible. Genome scan was performed using 137 polymorphic autosomal markers at an average spacing of 12 cM. Survival time and bacteremia were the phenotypic traits used for linkage assessment.

Significant linkage to survival time mapped to 9.4 cM on mouse Chromosome 7 with LOD=6.34 at D7Mit77. This locus is named Spir1 (Streptococcous pneumoniae infection resistance 1) and also shows suggestive linkage to bacteremia (LOD=4.62). BALB/cOlaHsd-derived alleles at Spir1 confer resistance to infection with S. pneumoniae with a dominant mode of inheritance while CBA/CaOlaHsd-derived alleles at Spir1 confer susceptibility with a recessive mode of inheritance. The 1.5 LOD support interval of Spir1 spans a 7 cM interval between D7Mit341a (8 cM) and D7Mit247 (16 cM). Potential candidate genes mapping to this interval are Tyrobp, Hcst, and Zfp36. The Spir1 interval is syntentic to human Chromosome 19q13.1-q13.3.
