Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 400 (C57BL/6J x DBA/2J)F2 animals and 26 BXD RI strains to identify QTLs associated with skeletal phenotypes. A panel of 672 microsatellite markers was used in the genome scan for the BXD RI group and 96 microsatellite markers at a resolution of 15-20 cM were used in the genome scan for the F2 group. Loci identified with significant linkage in the F2 group and confirmed in the BXD RI group are reported here.

On mouse Chromosome 1, a region from 60 cM - 100 cM showed linkage to tibia length, tibia medullary area, tibia CSMI, and femur length. The strongest association was at 74 cM for tibia length (LOD=10). This locus is named Bntq1 (bone traits QTL 1). The C57BL/6J allele at Bntq1 confers increased femur width, and increased tibial stiffness, tibial CSMI, tibia medullary area, and tibial sagittal width. Linkage to adiposity was also detected in this interval at 91 cM (LOD=6.1) with the DBA/2J-derived allele conferring increased adiposity. Bntq1 may exhibit a stronger effect in males for some bone traits. Previously identified bone QTLs mapping near Bntq1 are Bmd1 (87.9 cM), and Bmd5 (81.6 cM). A second locus distal to and overlapping with Bntq1 showed linkage to tibia medullary area at 93 cM with LOD=4.5. This locus isnamed Bntq2. The QTL interval of Bntq2 is from 72 cM - 101 cM. A previously identified bone QTL mapping near Bntq2 is Fembrs5 (101.5 cM).
