Analysis of interval specific congenic lines resolved the Idd16 diabetes QTL into two distinct loci named Idd16a and Idd16b. ALR/LtJ-derived portions of the Idd16 locus were introgressed onto the genetic background of NOD/LtJ. Parental strain ALR/LtJ is resistant to insulin-dependent (type 1) diabetes whereas parental strain NOD/LtJ is susceptible. Three different subcongenic lines were contructed.

Idd16a is localized to the distal portion of the original Idd16 QTL interval between D17Mit13 (19.06 cM) and H2-D (19.09 cM). The ALR/LtJ-derived allele of Idd16a appears to slow insulitis progression, but not its prevention. Potential candidate genes for Idd16a are Tnf (19.06 cM) and H2-Ddx (19 cM). The latter is considered a stronger candidate gene since serum levels of Tnf do not differ between NOD/LtJ and ALR/LtJ. The H2-Ddx allele is thought to be involved in resistance to type 1 diabetes.

Idd16b is localized to the proximal portion of the original Idd16 QTL interval above D17Mit16. Idd16b is involved in resistance to diabetogenesis.
