Mapping and Phenotype information for this QTL, its variants and associated markersJ:6610QTL associated with susceptibility to radiation-induced leukemia were mapped using a population of (A/J x C57BL/10ScSn)F2 intercross animals. Parental strain C57BL/10ScSn is susceptible to radiation-induced leukemia compared to parental strain A/J. (A/J x C57BL/10ScSn)F1 hybrids exhibit intermediate susceptibility. C57BL/10ScSn exhibits shorter latency and higher mortality than A/J. (A/J x C57BL/10ScSn)F1 hybrids exhibit longer latency similar to parental A/J but have high mortality similar to C57BL/10ScSn. Female animals appear to be more susceptible to leukemia induction but the difference was not found to be statistically significant. A major locus for resistance to radiation-induced leukemia mapped to mouse Chromosome 2 near Ly11 (69 cM). This locusis named Ril1 (radiation-induced leukemia sensitivity 1). A/J-derived alleles at Ril1 confer resistance to radiation-induced leukemia with codominant inheritance. The Ril1 locus is thought to be proximal to Ly11. A locus on mouse Chromosome 4 shows linkage to radiation-induced leukemia susceptibility near Cxv2 (62 cM). This locus is named Ril3 (radiation-induced leukemia sensitivity 2). Ril3 is estimated to be 1-3 cM proximal of Cxv2. F2 animals homozygous or heterozygous for C57BL/10ScSn-derived alleles at Ril3 exhibit increased survival after radiation exposure compared to animals homozygous for A/J-derived alleles at Ril3. Cxv2 is considered a likely candidate gene for Ril3 as Cxv2 expression is greater in susceptible strain A/J compared to resistantstrain C57BL/10ScSn.A C57BL/10ScSn-derived resistance locus mapped to mouse Chromosome 1 near Pep3 (71 cM). This locus is named Ril2 (radiation-induced leukemia sensitivity 3). C57BL/10ScSn-derived alleles at Ril2 (heterozygous or homozygous) confer increased survival after radiation exposure. Bxv1 (92.3 cM) maps near Ril2 and may be a possible candidate gene. |
