Mapping and Phenotype information for this QTL, its variants and associated markers

J:99478

Linkage analysis was performed on 396 animals from a (C57BL/6J x DBA/2J)F2 intercross and 22 BXD (B=C57BL/6J; D=DBA/2J) recombinant inbred (RI) strains to identify QTL associated with muscle weight variation at 500 days of age. 97 polymorphic markers at an average spacing of 15 cM - 20 cM were used for the genome scan. Parental strain C57BL/6J exhibits 13-22% greater muscle weight compared to parental strain DBA/2J.

On mouse Chromosome 1, linkage to soleus weight and tibialis anterior weight mapped to 102 cM near D1Mit221. This locus is named Skmw6 (skeletal muscle weight 6). The 1.5 LOD support interval of Skmw6 spans 89 cM - telomere. DBA/2J-derived alleles at Skmw6 confer increased muscle weight with a dominant mode of inheritance. Potential candidate genes for Skmw6 are Mrps9 (formerly 2310002A08Rik), Usp21 (92.6 cM), and Casq1 (94 cM).

On mouse Chromosome 2, three separate but overlapping QTL were identified. Skmw7 (skeletal muscle weight 7) maps to 20 cM near D2Mit323 and is linked to extensor digitorum longus (EDL) weight. The 1.5 LOD support interval of Skmw7 spans 0 cm - 65 cM. C57BL/6J-derived alleles at Skmw7 confer increased muscle weight with an additive mode of inheritance. Skmw8 (skeletal muscle weight 8) maps to 45 cM near D2Mit300 and is linked to tibialis anterior weight. The 1.5 LOD support interval of Skmw8 spans 37 cM - 62 cM. C57BL/6J-derived alleles at Skmw8 confer increased muscle weight with a recessive mode of inheritance. Skmw9 (skeletal muscle weight 9) maps to 62 cM near D2Mit300 and is linked to gastrocnemius muscle weight. The 1.5 LOD support interval of Skmw9 spans 12 cM - 82 cM. C57BL/6J-derived alleles at Skmw9 confer increased muscle weight with an additive mode of inheritance. A potential candidate gene for Skmw7, Skmw8, and Skmw9 is Capn3 (67.2 cM).

On mouse Chromosome 6, linkage to gastrocnemius muscle weight mapped to 21 cM near D6Mit188. This locus is named Skmw10 (skeletal muscle weight 10). The 1.5 LOD support interval of Skmw10 spans 0 cM - 57 cM. C57BL/6J-derived alleles at Skmw10 confer increased muscle weight with a dominant mode of inheritance. A potential candidate gene for Skmw10 is Clcn1 (22.5 cM).

On mouse Chromosome 9, linkage to soleus weight, tibialis anterior weight, and extensor digitorum longus(EDL) weight mapped to 61 cM near D9Mit212. This locus is named Skmw11 (skeletal muscle weight 11). The 1.5 LOD support interval of Skmw11 spans 51 cM - 69 cM. C57BL/6J-derived alleles at Skmw11 confer increased muscle weight with a dominant mode of inheritance. Potential candidate genes for Skmw11 are Mras and Myl3 (61 cM).
