Mapping and Phenotype information for this QTL, its variants and associated markersJ:97905Linkage analysis was performed on 361 female animals from the outbred strain DDY to identify loci associated with IgA nephropathy (IgAN). The genome scan utilized 270 microsatellite markers at an average spacing of 8 cM. Animals were grouped by onset of renal injury (early, late, or quiescent). The DDY strain exhibits significantly increased serum IgA and urinary albumin concentration compared to control strain BALB/c at all time points (20, 40, and 60 weeks of age). Linkage to early onset renal injury mapped to 87.2 cM on mouse Chromosome 1 near D1Mit16. This locus also shows suggestive linkage to late onset renal injury. The QTL symbol Igan2 has been assigned for this locus. The Igan2 interval is flanked by markers D1Mit268 (83.4 cM) and D1Mit112 (91.3 cM). A potential candidate gene is Sele at 86.6 cM. Linkage to early and late onset renal injury mapped to 17 cM on mouse Chromosome 9 near D9Mit252. The QTL symbol Igan3 has been assigned for this locus. The Igan3 interval is flanked by markers D9Mit88 (12 cM) and D9Mit285 (21 cM).Linkage to early onset renal injury mapped to 17 cM on mouse Chromosome 10 near D10Mit86. This locus also shows suggestive linkage to late onset renal injury. The region of peak linkage is syntenic to a locus on human Chromosome 6q22-q23 named IGAN1 that is associated with familial susceptibility to IgAN. Since the mouse locus is syntenic to the human locus MGI has assigned Igan1 for the mouse QTL at D10Mit86. The Igan1 interval is flanked by markers D10Mit50 (7 cM) and D10Mit85 (22 cM).Linkage to serum IgA concentration mapped to 58 cM on mouse Chromosome 12 near D12Mit20. This locus is designated Igan4. The Igan4 QTL interval is flanked by markers D12Mit16 (53 cM) and D12Mit144 (61 cM). A potential candidate gene mapping near Igan4 is Igh (58 cM). |
