Linkage analysis was performed on a series of AXB/BXA (A=A/J; B=C57BL/6J) recombinant inbred (RI) strains and 17 AcB and 23 BcA recombinant congenic (RC) strains to identify QTL associated with voluntary alcohol consumption (VAC). Animals were phenotypedat 8 weeks of age. After 4 days of forced exposure to 10% alcohol solution, animals were given free choice between water and 10% alcohol solution for 3 weeks. A panel of 780 polymorphic markers was used for the genome scan.

In the AXB/BXA RI series, significant linkage to alcohol preference mapped to 107 cM on mouse Chromosome 2 near D2Mit74 (LOD=6.13). This locus accounts for 30% of the variance in male and female mice and is named Vacq1 (voluntary alcohol consumption QTL 1). The Vacq1 QTL interval is defined by markers D2Mit148 (105 cM) and D2Mit266 (109 cM). C57BL/6J-derived alleles at Vacq1 confer increased alcohol consumption. Potential candidate genes for Vacq1 are Ntsr1 (107 cM) and Chrna4 (108 cM). Vacq1 appears to be influenced by a locus on mouse Chromosome 15 named Vacq2 (voluntary alcohol consumption QTL 2). Animals homozygous for C57BL/6J-derived alleles at both Vacq1 and Vacq2 exhibit significantly increased alcohol preference.

Vacq2 on mouse Chromosome 15 is linked to Pdgfb (47 cM). Potential candidate genes for Vacq2 are Bzrp (43.3 cM), and Cyp2d (47.2 cM). Previously identified QTL mapping near Vacq2 are Drb7 (45 cM) and Lore5 (46 cM).

Significant linkage to alcohol preference in male animals mapped to 5 cM near D2Mit359 in the AcB RC series. This locus has a male-specific effect and is named Vacq3 (voluntary alcohol consumption QTL 3). The Vacq3 QTL interval spans 5 cM - 31.7 cM. C57BL/6J-derived alleles at Vacq3 confer increased alcohol consumption. Previously identified alcohol preference QTL mapping near Vacq3 are Etohc1 (28 cM) and Etohcta2 (23.5 cM). Significant linkage to alcohol preference in male and female animals mapped to 30 cM on mouse Chromosome 2 near D2Mit241 in the BcA RC series. This locus is named Vacq4 (voluntaryalcohol consumption QTL 4). The Vacq4 QTL interval spans 30 cM - 37 cM. A/J-derived alleles at Vacq4 confer increased alcohol consumption. Previously identified QTL mapping near Vacq4 are Ap2q (34 cM), Alcp1 (47.5 cM), and Alcw4 (37 cM).

Significant linkage to alcohol preference in male animals of the AcB RC series mapped to 16 cM on mouse Chromosome 13 near D13Mit266. This locus is named Vacq5 (voluntary alcohol consumption QTL 5). The QTL interval of Vacq5 spans 9 cM - 30 cM. C57BL/6J-derived alleles at Vacq5 confer increased alcohol consumption in male animals. A potential candidate gene for Vacq5 is Agtr1 (16 cM).

Significant linkage to alcohol preference mapped to 4 cM on mouse Chromosome 10 near D10Mit80 in the BcA RC series. This locus is named Vacq6 (voluntary alcohol consumption QTL 6), and the QTL interval spans 4 cM - 5 cM. A/J-derived alleles at Vacq6 confer increased alcohol consumption in male and female animals.

Significant linkage to alcohol preference mapped to 1 cM on mouse Chromosome 12 near D12Mit37 in the BcA RC series. This locus is named Vacq7 (voluntary alcohol consumption QTL 7). The Vacq7 QTL interval spans 1 cM - 5 cM. A/J-derived alleles at Vacq7 confer increased alcohol consumption in male and female animals.

In the AXB/BXA RI series, suggestive linkage to alcohol preference mapped to 8 cM on mouse Chromosome 11 near Pmv22, 21 cM on mouse Chromosome 10 near D10Mit126, 1 cM on mouse Chromosome 12 near D12Mit37, and to 29 cM on mouse Chromosome 16 near D16Mit125. A suggestive male-specific locus mapped to 63.5 cM on mouse Chromosome 7 near D7Mit105, and a suggestive female-specific locus mapped to 47.5 cM on mouse Chromosome 11 near D11Mit120.

In the AcB RC series, suggestive linkage to alcohol preference mapped to 15 cM on mouse Chromosome 5 near D5Mit387, 63.5 cM on mouse Chromosome 7 near D7Mit105, 19 cM on mouse Chromosome 8 near D8Mit292, 3 cM on mouse Chromosome 10 near D10Mit298, and to 59.2 cM on mouse Chromosome 15 near D15Mit42. In the BcA RC series, suggestive linkage to alcohol preference mapped to 68 cM on mouse Chromosome 9 near D9Mit281.
