Linkage analysis was performed on 276 (BALB/cOla x C57BL/6Ola)F1 x BALB/cOla backcross animals to identify QTL associated with T-cell secretion of IL-4. CD4+ T-cells from 6- to 8-week old backcross animal were evaluated in vitro. Parental strain BALB/cOla exhibits high IL-4 secretion (~900 pg/ml) whereas parental strain C57BL/6Ola exhibits low IL-4 secretion (<50pg/ml). 83 polymorphic markers at an average spacing of 20 cM were used for the genome scan (94% coverage). Three novel QTL (Tsiq1, Tsiq2, andTsiq3) with additive effects on IL-4 secretion were identified in this study. Animals inheriting high alleles at all 3 loci exhibit significantly increased IL-4 secretion, but not as high as BALB/cOla parentals.

Significant linkage to T-cell secretion of IL-4 mapped to mouse Chromosome 7 between D7Mit66 (57.5 cM) and D7Mit206 (60 cM). This locus explains 24% of the variance and is named Tsiq1 (T-cell secretion of IL-4 QTL 1). Homozygosity for BALB/cOla-derived alleles at Tsiq1 confer high IL-4 secretion. Apreviously identified IL-4 commitment QTL named Dice2 (63.5 cM) maps near Tsiq1. Representational difference analysis (RDA) was used to screen candidate genes for Tsiq1. Thumpd1 (Flj20274) maps within the Tsiq1 peak and exhibits differential expression between BALB/cOla and C57BL/6Ola T-cells. A 349 bp deletion in the BALB/cOla allele may be the cause of Thump1 overexpression in BALB/cOla activated T-cells.

Tsiq2 (T-cell secretion of IL-4 QTL 2) mapped to mouse Chromosome 19 near D19Mit46 (24 cM) and D19Mit53 (43 cM). This locus explains 20.4% of the variance. Homozygosity for BALB/cOla-derived alleles at Tsiq2 confer high IL-4 secretion. Potential candidate genes for Tsiq2 are Jak2 (24 cM), Za20d2 (15 cM; formerly Zfp216), and Smarca2 (17 cM).

Tsiq3 (T-cell secretion of IL-4 QTL 3) mapped to 52.2 cM on mouse Chromosome 15 near D15Mit34. This locus explains 25.3% of the variance. A C57BL/6Ola-derived allele (heterozygous) at Tsiq3 confers high IL-4 secretion. Potential candidate genes for Tsiq3 are Ptk2 (42 cM) and Sox10 (46.6 cM).
