Mutation detailsA targeting construct was designed to insert Cys322Thr and Cys323Ser modifications in the locus to eliminate both palmitoylation sites. A tACE-Cre-neo cassette was included in the construct and self-excised upon passage through the male germ line. Expression of palmitoylation-deficient rhodopsin transcript was demonstrated by RT-PCR, and the absence of palmitates on rhodopsin protein of mutant mice was confirmed by mass spectrometry.
