Mutation details The transgene consists of the entire Prdx6 sequence (approximately 10 kB) and 6.6 kB of upstream sequence from the diet-induced atherosclerosis resistant 129X1/SvJ mouse strain. The 129X1/SvJ sequence contains an amino acid variant at position 124 encoding aspartic acid rather than the alanine found in C57BL/6. Southern blot analysis distinguishes between endogenous and transgenic genes. Lines 153 and 105 contained multiple copies of the transgene and were selected for further analysis. Western blot, ELISA and immunohistochemical analysis reveal higher levels of PRDX6 expression in many tissues, especially liver and aorta, than is found in tissues normally expressing the gene.
