Cia5, an arthritis susceptibility QTL on mouse Chromosome 3, was resolved into 3 separate loci (Cia5a, Cia5b, and Cia5c) using congenic line analysis. Congenic and subcongenic animals carrying portions of the Cia5 locus derived from resistant strain RIIIS/J on a susceptible B10.RIII background were created.

Cia5a maps between D3Mit187 (42.4 cM) and D3Mit49 (44.8 cM) on mouse Chromosome 3. Interaction between Cia5a and a locus on Chromosome 15 named Cia31 (14.5 cM) affects onset and early arthritis severity in female animals (LOD=6.1). Heterozygosity at Cia31 and RIIIS/J-derived alleles at Cia5a confers delayed onset of collagen-induced arthritis in female animals with an additive effect. Interaction between Cia5a and Cia32 (17.8 cM on mouse Chromosome 15) affects arthritis severity in the early (LOD=12.1) and late phases of disease (14.5 cM) in both male and female animals. Interaction between Cia5a and Cia30 (9.9 cM on mouse Chromosome 15) affects arthritis severity in the late phase of disease (LOD=11.8) in both male and female animals. A previously identified diabetes QTL named Idd17 (39 cM) maps near Cia5a.

Cia5b maps between D3Mit75 (49 cM) and D3Mit284 (51.1 cM). RIIIS/J-derived alleles at Cia21 confers reduced late onset arthritis severity in female animals. Cia5b interacts epistatically with a locus on mouse Chromosome 15 named Cia32 (LOD=13.6). Heterozygosity at Cia32 and RIIS/J-derived alleles at Cia21 decreases arthritis score in female animals with a dominant effect. A previously identified diabetes QTL named Idd10 (48.5 cM) maps near Cia5b.

Cia5c maps between D3Mit103 (51.1 cM) and D3Mit370 (43.8 cM). RIIIS/J-derived alleles at Cia5c confers reduced late onset arthritis severity in female animals. Cia5c also interacts with Cia32 on mouseChromosome 15. Additive interaction between Cia5c and Cia5a affects arthritis severity in female animals (LOD=6.1). A previously identified diabetes QTL named Idd18 (53.3 cM) maps near Cia5a.
