Mapping and Phenotype information for this QTL, its variants and associated markers

Loci associated with susceptibility to bleomycin-induced pulmonary fibrosis were mapped using 36 recombinant congenic (RC) strains derived from C57BL/6J (susceptible) and A/J (resistant). Three to five animals of each recombinant congenic strain were exposed to bleomycin and phenotyped for lung fibrosis 3 weeks later.

Among the BcA (C57BL/6J x A/J) RC strains, significant linkage to bleomycin-induced pulmonary fibrosis susceptibility mapped to mouse Chromosomes 9, 15, and 18. These loci are described below. Among all RC strains tested, suggestive linkage to bleomycin-induced pulmonary fibrosis susceptibility mapped to mouse Chromosomes 1, 3, 5, 6, 9, and 12. C57BL/6J-derived alleles confer susceptibility to bleomycin-induced pulmonary fibrosis all suggestive loci, many of which are male-specific. Potential candidate genes were identified for suggestive QTL on mouse Chromosomes 1, 5, 6, and 12 using gene expression analysis of lung mRNA from bleomycin-treated A/J and C57BL/6J animals. (This data can be found on Table 4 of the publication.)

Blmpf3 (bleomycin-induced pulmonary fibrosis 3) mapped to 43 cM on mouse Chromosome 9 near D9Mit236 (LOD=4.92). A/J-derived alleles at Blmpf3 confer susceptibility to bleomycin-induced pulmonary fibrosis.

Blmpf4 (bleomycin-induced pulmonary fibrosis 4) mapped to 55.6 cM on mouse Chromosome 15 near D15Mit108 (LOD=3.51). A/J-derived alleles at Blmpf4 confer susceptibility to bleomycin-induced pulmonary fibrosis. Igta5 (57.4 cM), Irak4, and BC031593 are candidate genesidentified by gene expression analysis. Igta5 and Irak4 exhibit decreased expression in bleomycin-treated A/J animals, and BC031593 exhibits increased expression in lungs of bleomycin-treated A/J animals, compared to bleomycin-treated C57BL/6J.

Blmpf5 (bleomycin-induced pulmonary fibrosis 5) mapped to 37 cM on mouse Chromosome 18 near D18Mit183 (LOD=3.51). A/J-derived alleles at Blmpf5 confer susceptibility to bleomycin-induced pulmonary fibrosis. Lox (29 cM) and Iigp1 are candidate genes identified by gene expression analysis. Lox and Iigp1 expression both decreased in lungs of bleomycin-treated A/J animals, compared to bleomycin-treated C57BL/6J.
