Mapping and Phenotype information for this QTL, its variants and associated markersJ:101740Previously identified diabetes QTL Idd21 on mouse Chromosome 18 was further localized in this experiment. The diabetes resistant NOD-Chr 18ABH consomic was used to generate subcongenic lines carrying different ABH-derived portions of chromosome 18. Analysis of subcongenic lines resolved Idd21 into 3 separate loci- Idd21a and Idd21b on distal chromosome 18 and Idd21c on proximal chromosome 18. Idd21a and Idd21b exhibit orthology to human and rat diabetes susceptibility loci IDDM6 (18q12-q21) and Iddm3, respectively. Idd21c exhibits orthology to a human diabetes locus named IDDM10 (10p11-q11). The Idd21a interval maps to the distal 18.1 Mb portion of mouse Chromosome 18 and spans approximately D18Mit8 (47 cM) to D18Mit4 (57 cM). ABH-derived alleles at Idd21a confer diabetes resistance. Potential candidate genes for Idd21a include Smad2 (48 cM), Smad7, and Smad4 (48 cM). Idd21b maps proximal to Idd21a and spans approximately 61.7 Mb to 75 Mb. The flanking markers for Idd21b are D18Mit51 (37 cM) and D18Mit8 (47 cM). ABH-derived alleles at Idd21b confer diabetes susceptibility. Potential candidate genes for Idd21a include Smad4 (48 cM), Dcc (45 cM), and Tcf4.Idd21c maps to the proximal 21.9 Mb portion of mouse Chromsome 18 between D18Mit64 (2 cM) and D18Mit111 (11 cM). ABH-derived alleles at Idd21c confer diabetes resistance. No potential candidate genes for Idd21c were identified in this study. |
