Mapping and Phenotype information for this QTL, its variants and associated markersJ:103483Linkage analysis was performed on 492 (DBA/1J x 129X1/SvJ)F2 animals to identify loci associated with ear punch wound healing. Parental strain DBA/2J is a good healer and parental strain 129X1/SvJ is a poor healer. Interestingly, F1 animals exhibit woundhealing similar to or worse than the poor healer parental strain 129X1/SvJ. Genome scan was conducted with 98 polymorphic microsatellite markers. Ear hole wounds were measured at 3, 7, 15, 21, and 25 days after the ear punch was made. Four ear healing QTLaccounting for 17.1% of the phenotypic variance were identified in this study. Several minor interacting loci (LOD<2.7) accounting for 55.7% of the variance were also detected. For a complete list of minor interacting loci please refer to Table 2 of thisreference (J:103483) by Masinde et al, 2005. Earheal1 mapped to 101.2 cM on mouse Chromosome 1 near D1Mit406 (LOD=4.1). This locus explains 6.3% of the phenotypic variance. 129X1/SvJ-derived alleles at Earheal1 confer increased wound healing with recessive inheritance.Earheal2 mapped to 66.6 cM (137.5 Mb) on mouse Chromosome 4 near D4Mit170 (LOD=2.9). This locus explains 4.3% of the phenotypic variance. Heterozygosity for DBA/1J and 129X1/SvJ alleles at Earheal2 appear to confer increased wound healing. The mode of inheritance is unclear. Previously identified wound healing QTL Stheal4 (51.3 cM) maps near Earheal2. Potential candidate genes for Earheal2 include Ephb2 (65.7 cM; 135.72 Mb), Epha2 (73.2 cM; 140.21 Mb), and Epha8 (135.84 Mb).Earheal3 mappedto 45 cM (86.93 Mb) on mouse Chromosome 12 near D12Mit118 (LOD=3.1). This locus explains 2.9% of the phenotypic variance. DBA/1J-derived alleles at Earheal3 confer increased wound healing with recessive inheritance. Adam4 (78.28 Mb) and Adam21 (28 cM;78.42 cM) are potential candidate genes for Earheal3.Earheal4 mapped to 17 cM (36.11 Mb) on mouse Chromosome 18 near D18Mit12 (LOD=3.2). This locus explains 3.6% of the phenotypic variance. DBA/1J-derived alleles at Earheal4 confer increased wound healingwith recessive inheritance. Potential candidate genes for Earheal4 include Pcdh12 (38.49 Mb), the protocadherin alpha gene cluster (37.15 Mb), the protocadherin beta gene cluster (37.49 Mb - 37.74 Mb), and the protocadherin gamma gene cluster (38.07 Mb). |
