Mapping and Phenotype information for this QTL, its variants and associated markers

Anosmia to isovaleric acid was mapped to mouse Chromosome 4 using a series of BXD (B=C57BL/6J; D=DBA/2J) recombinant inbred (RI) strains. Parental strain C57BL/6J exhibits specific anosmia to isovaleric acid whereas parental strain DBA/2J is osmic. A panel of 900 polymorphic markers was used to analyze strain distribution patterns in the RI set. Iva1 (isovaleric acid smelling 1) mapped to 56.7 cM on mouse Chromosome 4 between D4Mit37 (56.5 cM) and D4Mit249 (58.2 cM). A suggestive locus named Iva2 mapped to 69.5 cM on mouse Chromosome 6 near D6Mit15 (74 cM, LOD=1.7).

(C57BL/6J x DBA/2J)F1 x C57BL/6J backcross animals were used to test the allelic effects of Iva1 and Iva2. Animals homozygous for C57BL/6J-derived alleles at both Iva1 and Iva2 were anosmic to isovaleric acid. Neither locus alone is thought to be sufficient for isovaleric-specific osmia.

Fine mapping was performed using 734 backcross animals. Iva1 was resolved to a 0.27 cM interval between D4Mit37 (56.5 cM) and D4Mit156 (56.8 cM). Iva2 was localized to a 7.5 cM interval between D6Mit198 (67 cM) and D6Mit137 (71.8 cM).
