Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 328 (MRL/MpJ x CAST/EiJ)F2 intercross animals to identify loci associated with soft tissue regeneration. Genome scan was conducted with 86 polymorphic markers paced approximately 17 cM apart. Soft tissue regeneration wasassessed by evaluation of ear hole wound healing at regular time intervals up to 25 days after administration of the ear punch. Parental strain MRL/MpJ is a fast healer whereas parental strain CAST/EiJ is a poor healer.

A locus associated with ear woundhealing at all time points mapped to 66.6 cM on mouse Chromosome 4 to D4Mit170 with the highest statistical significance, LOD=3.85, for ear wound healing at day 15. This locus is thought to be the same as the previously identified Stheal4 (soft tissue heal4).

03.05.2015 Curators Note: Stheal4 was originally mapped in J:73197 in 2001 using a (MRL/MpJ x SJL/J)F2 cross, which differs from the cross used here, we consisder this a separate map study and have named the QTL Stheal12. Speculation is also offered that this QTL may be the same as Heal8. Heal8 now maps to mouse Chr 13 in MGI, mapped in J:50111 in 1998 using an (MRL/MpJ-Faslpr x C57BL/6NTac)F2 intercross.

MRL/MpJ-derived alleles at Stheal12 confer increased wound healing with a dominant mode of inheritance. This locus explains 6.9% of the phenotypic variance. Stheal12 interacts with Stheal11 on mouse Chromosome 17 (see below). Previously identified QTL Earheal2 (66.6 cM) overlaps with Stheal12.

A broad region spanning 26 cM - 61 cM on mouse Chromosome 9 showed linkage to ear wound healing. The proximal portion of this region around 26 cM (D9Mit129) is thought to correspond to Stheal8, and the distal portion of this region around 49 cM - 61 cM is thought to correspond to Stheal9.

03.05.2015 Curators Note: Steal8 and Steal9 were also originally mapped in J:73197 in 2001 using a (MRL/MpJ x SJL/J)F2 cross, which differs from the cross used here. We consider this a separate map study and have named the QTL identified here as Stheal13 and Stheal14 respectively.

The proximal QTL; mapping to 26 cM with marker D9Mit129 on mouse Chromosome 9, LOD=2.2; Stheal13 accounts for 3.7% of the phenotypic variance, with the MRL/MpJ derived allele conferring increased healing.

Maximum linkage for Stheal14 occursat 49 cM with D9Mit198 (LOD=3.26) and 53 cM with D9Mit355 (LOD=3.27). CAST/EiJ-derived alleles at 49 cM confer increased wound healing and explains for 9.8% of the variance, whereas MRL/MpJ-derived alleles at 53 cM confer increased wound healing and explains 5.5% of the variance. Authors speculate it unlikely that two closely linked loci reside within this short region.

Linkage to ear wound healing mapped to 22.9 cM on mouse Chromosome17 with D17Mit51. The highest statistical significance was LOD=5.13 for wound healing at day 25. This locus explains 8.3% of the phenotypic variance and is designated Stheal11 (soft tissue heal 11). CAST/EiJ-derived alleles at Stheal11 confer increasedwound healing with a dominant mode of inheritance.

There is evidence of interaction between Stheal11 and Stheal12, and between Stheal11 and Stheal14. Homozygosity for MRL/MpJ-derived alleles at Stheal12 or Stheal14 in conjunction with homozygosity for CAST/EiJ-derived alleles at Stheal11 is associated with the best woundhealing.

Suggestive linkage to ear wound healing mapped to 5cM on mouse Chromosome 1 near D1Mit64 (LOD=2.13) and to 53.3 cM on mouse Chromosome 7 near D7Mit98 (LOD=2.31).MRL/MpJ-derivedalleles are associated with increased wound healing at these suggestive loci.
