Mutation detailsCre mediated recombination removed a floxed neo, leaving exon 1, containing the ATG, replaced by a c-Myc epitope tagged Epas1 cDNA such that knock-in expression depended upon the endogenous regulatory sequence. Mutant ES cells did not express Hif1a protein, as shown by Western blot and EMSA analyses. Immunoprecipitation and immunoblot detected expression of the knock-in sequence. (J:97164)

Notes
Injection of homozygous ES cells into nude mice promotes faster growth of larger, more hemorrhagic tumors than wild-type ES cells. These tumors have increased microvessel density and a wider range of differentiated cell types than tumors from wild-type cells (J:97164). Homozygous ES clones form significantly fewer CFUs (hematopoietic colony-forming units) of all hematopoietic lineages and form 7.8-fold more secondary embryoid bodies (Ebs) compared with wild-type, consistent with maintenance of an undifferentiated state (J:105812).
