Mapping and Phenotype information for this QTL, its variants and associated markersJ:3240Confirmation of linkage between H2-D and Rmp3 on mouse Chromosome 17 was established. (C57BL/6NCr x DBA/2NCr)F1 female mice were backcrossed to DBA/2NCr male mice deriving an N2 founder male of the genotype H2-Db Rmp3r with the C57BL/6 allele H-2Dbbeing a marker for the Rmp3 resistant phenotype. Data derived from the concordant passage of these two genes through seven generations of backcrosses to DBA/2NCr suggest a recombination frequency of 0.2 to 0.3 between H2-D and Rmp3. 10.8.2015 Curator Note: Rmp3 was orginally mapped using a (C57BL/6J x DBA/2J)F1 x DBA/2J backcross in 1991 in J:11032. A second cross in the same study identified QTL Rmp6 using a (C57BL/6NCr x DBA/2NCr)F1 x DBA/NCr backcross population. We have equated the map study here as confirming Rmp6.J:11032Linkage to mousepox susceptibility was mapped in 101 male animals from a (C57BL/6J x DBA/2J)F1 x DBA/2J backcross inoculated with 105 PFU of mouse ectromelia virus. Inbred strain DBA/2J is highly susceptible to infection with ectromelia virus whereas inbred strain C57BL/6J is resistant. Rmp2 (resistance to mousepox 2) and Rmp3 (resistance to mousepox 3) were identified near Hc (23.5 cM) on mouse Chromosome 2 and near H2-D (19 cM) on mouse Chromosome 17, respectively. Homozygosity for DBA/2J-derived alleles at Rmp2 and Rmp3 is associated with susceptibility to ectromelia virus infection and increased post-infection mortality. 10.8.2015 Curator Note: When previoulsy identified QTL are cited as confirmed or verified within the same study, using a different cross then that initially used, we have created a new QTL. We consider each distinct cross to be a separate mapping experiment. We have named the QTL identified here using the (C57BL/6NCr x DBA/2NCr)F1 x DBA/NCr backcross as Rmp5 and Rmp6 repectively.Analysis of 66 males animals from a (C57BL/6NCr x DBA/2NCr)F1 x DBA/NCr backcross had similar results. When the experiment was repeated on gonadectomized and sham-operated male backcross animals, Rmp5 and Rmp6 were shown to confer additional protection against ectromelia virus in sham-operated animals. This finding indicates that Rmp5 and Rmp6 exhibit responsiveness to hormonal sexfactors. The complement component C3 (34.3 cM) maps near Rmp6. C3 is androgen-responsive and may be a possible candidate gene. |
