Mapping and Phenotype information for this QTL, its variants and associated markersJ:88524Genetic loci confering susceptibility to repetitive pinch-induced catalepsy were mapped in 296 CBA/Nov x (CBA/Nov x AKR/J)F1 backcross animals. 65 polymorphic markers for the genome scan. Parental strain CBA/Nov is susceptible to pinch-induced catalepsy (lasting longer than 1 minute) compared to parental strain AKR/J. Significant linkage to catalepsy susceptibility mapped to 74 cM on mouse Chromosome 13 near D13Mit78 (P<0.0016). This locus is named Catys1 (catalepsy susceptibility 1). CBA/Nov-derived alleles at Catys1 confer susceptibility to pinch-induced catalepsy as 74% of backcross animals homozygous for this allele exhibit the phenotype. Potential candidate genes for Catys1 include Itga1 (65 cM), Itga2 (64 cM), Fgf10 (75 cM), and Htr1a (58 cM).Suggestive linkage to catalepsy susceptibility mapped to 61 cM on mouse Chromosome 13 near D13Mit76 (P=0.058).J:149041QTL mapping using congenic lines in this experiment confirmed the location of Catys1 to mouse Chromosome 13 (J:88524) and further refined its location to a 10 cM region. CBA/lacJ are catalepsy-prone whereas AKR/J mice are catalepsy resistant. Three congenic mouse lines (AKR.CBAD13Mit76C,AKR.CBA-D13Mit76A and AKR.CBA-D13Mit78)carrying from between 59 and 75 cM fragments from mouse Chromosome 13 derived from the CBA to the AKR genome were created via the backcrossing of (CBA X AKR)F1 on AKR strain. These aformantioned Congenic lines proved informative in mapping catalepsy to mose Chromosome 13. In a genome wide scan using 48 markers polymorphic between AKR/J and CBA/lacJ the Catys1 locus was confirmed to be in mouse Chromosome 13 between 61 and 70- cMand associated near D13Mit78. The Chromosome 13 gene accounted for 20% of the trait penetrance. |
