Mutation detailsThe enhanced green fluorescent protein (EGFP) coding sequence, followed by a termination codon, a polyadenylation signal and a loxP-flanked MC1-neo cassette, was inserted into the gene immediately following and in frame with the translation initiation codon in the second exon. The neo cassette was subsequently deleted by cre recombinase. EGFP is expressed under control of the native promoter. Absence of expression of the endogenous gene was demonstrated by Northern blot analysis, using a Slc39a1 cDNA probe, of total RNA and immunoblot analysis, using a polyclonal rabbit antiserum to a SLC38A1 peptide, of membrane proteins isolated from small intestine, kidney and liver of homozygous mutant mice. Subjects and controls were day E14 embryos from homozygous mutant or wild-type matings whose dams were fed a zinc-deficient diet from day 8 of pregnancy.

Double mutant subjects were day E14 embryos from matings of mice homozygous for both mutations whose dams were fed a zinc-deficient diet from day 8 of pregnancy.
