Mutation details: This allele is derived from Pik3ca and was produced as follows: A point mutation was engineered into the locus, resulting in the DFG to AFG conversion in the conserved DFG motif at the C-terminal kinase domain. A C-terminal Myc-epitope tag was introduced immediately preceding the translational stop codon as well as a floxed IRES-lacZ-geo cassette in the 3' UTR. The floxed sequence was subsequently removed by crossing with mice carrying the Tg(CMV-cre)1Cgn transgene. The p110alpha, p110beta, and p110gamma subunit expression was not altered in homozygous embryos and in liver, muscle and fat from hetrozygous mice. The heterozygous mutant demonstrated 50% loss of function in overall class IA PI(3)K activity in liver, muscle and fat.
