Mutation detailsA 69 bp fragment containing an AU rich element was inserted into the 3' untranslated region downstream of the stop codon via homologous recombination. In addition a self-excising neo resistance cassette consisting of a floxed mouse testes-specific Ace promoter fused to the cre recombinase and neomycin resistance genes was also inserted and then removed during germline transmission. RT-PCR showed that message levels are reduced about 40 - 50% in adipose tissue, liver, kidney, and heart in homozygotes compared to wild-type mice. Mice generated from one targeted ES cell clone (YTC-1) also have a tightly linked mutation Atp2b2m1Mae. (J:110607)
