Mutation detailsThe transgenic mouse line designated A8 was created using construct 3'1, which contains the 5' proximal promoter region of the Nodal locus extending from 5kb to 50 bp upstream of the translation initiation codon followed by the bacterial beta-galactosidase (lacZ) gene and a 3.5-kb segment of Nodal including intron 1, wherein resides a left side-specific asymmetric enhancer (ASE). Expression is observed throughout the epiblast of the embryo at E5.5, restricted to the proximal epiblast at E6.5, and in the anterior visceral endoderm from embryonic day E6.0 through E6.5. At the early somite stage, expression occurs on the left side of the lateral plate mesoderm (LPM) in the splanchopleure and somatopleure. (J:56146)
