Linkage analysis was performed on female animals from 13 CXB (C=BALB/cByJ; B=C57BL/6ByJ) recombinant inbred (RI) strains to identify QTLs associated with susceptibility to adenovirus induced hyperthyroidism. Animals between 5-8 weeks of age were injectedat regular intervals with adenovirus expressing the TSH receptor A-subunit. Serum T4, TSHR antibodies (measured by ELISA or inhibition of TSH binding), and thyroid histology were examined. After adenovirus injections, parental strain BALB/cJ is susceptible to hyperthyroidism and exhibits elevated serum T4 compared to parental strain C57BL/6J. A panel of 1384 markers was screened for QTL detection. Loci reaching statistical significance LOD>3 are assigned QTL symbols.

On mouse Chromosome 3, linkage to change in serum T4 after 2 adenovirus injections mapped to 28 Mb- 29 Mb with LOD=2.43.

On mouse Chromosome 10, linkage to serum T4 (LOD=1.50) and change in serum T4 after 3 adenovirus injections (LOD=2.56) mapped to approximately 62 cM (113.886 Mb - 114.056 Mb) near D10Mit233. TRH-degrading enzyme, Trhde (114 Mb), maps near this locus.

On mouse Chromosome 13, linkage to serum T4 before adenovirus injection mapped to 98.5 Mb - 99.3 Mb with LOD=2.56.

On mouse Chromosome 17, linkage to TSHR antibody production after 2 adenovirus injections mapped to 32.7 Mb - 36.9 Mb with LOD=3.57. This locus is named Advh1 (adenovirus induced hyperthyroidism 1). The H2 (23 cM; 31.96 Mb) locus maps near Advh1. A second locus named Advh2 (adenovirus induced hyperthyroidism 2) mapped to 45.24 Mb - 46.97 Mb near D17Mit66 (24.5 cM; LOD=3.57).

On mouse Chromosome 19, linkage to serum T4 after 2 adenovirus injections mapped to 43.7 Mb (LOD=2.70), and linkage to serum T4 after 3 injections mapped to 22.8 Mb - 24.58 Mb near D19Mit40 (25 cM; LOD=2.01).

On mouse Chromosome X, linkage to TSHR antibody production after 2 adenovirus injections mapped to 70.9 Mb - 81.6 Mb (LOD=3.57). This locus is named Advh3 (adenovirus induced hyperthyroidism 3). A second locus named Advh4 (adenovirus induced hyperthyroidism 4) mapped to 33.4 Mb - 43.2 Mb (LOD=3.24). Lamp2 (13 cM; 32.925 Mb) and Pdcd8 (17 cM; 42.995 Mb) map near Advh4.
