Mapping and Phenotype information for this QTL, its variants and associated markersJ:114560Linkage analysis was performed on 75 LXS recombinant inbred (RI) strains to identify QTLs associated with low dose ethanol-induced activation. Animals aged 55 to 85 days were tested in 3 independent cohorts using a 3-day protocol. A panel of 330 polymorphic loci at a resolution of 4.5 cM was genotyped in 648 experimental animals. Phenotypes measured were Ethanol-Saline (ES) total distance traveled and Ethanol-Baseline (EB) total distance traveled. Several suggestive loci were detected for both phenotypes, many of which map near suggestive ethanol-related loci identified by other investigators. The strongest linkage detected was at 19.1 cM (34.1 Mb) on mouse Chromosome 7 (LOD=3.04). This locus is associated with the ES phenotype in males and females. The QTL interval spans 21.8 cM to 25.2 cM.Suggestive linkage to the ES phenotype in males and females was also detected on mouse Chromosomes 12 (at 16.3 cM) and 13 (at 53.3 cM). SNP analysis confirmed these loci and also detected a suggestive female-specific locus at 72.1 cM on mouse Chromosome 8 (LOD=1.66). This locus is also linked to the EB phenotype in males and females (LOD=2.04). Suggestive linkage to the EB phenotype in males was detected at 73.1 cM on mouse Chromosome 3 (LOD=2.79). Novel QTLs Elda3 (ethanol induced low dose activation 3) and Elda4 mapped to mouse Chromosome 2 and 4, respectively. These loci are female-specific. Elda3 is associated with EB phenotype and mapped to 65.2 cM (127.8 Mb) on mouse Chromosome 2 near D2Mit304 (LOD=2.3). The QTL interval spans 23.8 cM to 33.2 cM. LS-derived alleles at Elda3 confer increased ethanol-induced locomotor activation in females. Elda4 is associated with EB and ES phenotypes and mapped to 20.4 cM (46.2 Mb) on mouse Chromosome 4 near D4Mit89 (LOD=1.87). The QTL interval spans 15.8 cM to 30.9 cM. SS-derived alleles at Elda4 confer increased ethanol-induced locomotor activation in females, whereas LS-derived alleles confer decreased activation in females. |
