Linkage analysis was performed on 284 animals from a (C57BL/6JOla x SWR/Ola)F2 intercross to identify QTL associated with non-heme iron levels in the liver and spleen. Animals were sacrificed and phenotyped at 8 weeks of age. Parental strain SWR/Ola displays high levels of liver and spleen non-heme iron compared to parental strain C57BL/6JOla.

Highly significant linkage to liver iron mapped to 57 cM on mouse Chromosome 2 near D2Mit17 (LOD=5.2). This locus also shows suggestive linkage to spleen iron content in males and is named Nhil1 (non-heme iron levels 1). The Nhil1 confidence interval spans 51 cM - 68 cM. C57BL/6JOla-derived alleles at Nhil1 confer increased liver and spleen iron. B2m (69 cM) is a potential candidate gene for Nhil1 and exhibits increased expression in C57BL/6JOla compared to SWR/Ola.

Highly significant linkage to liver iron mapped to 29 cM on mouse Chromosome 16 near D16Mit30 (LOD=8.1). This locus is also significantly linked to liver iron in the male subset (LOD=5.6) and is namedNhil2 (non-heme iron levels 2). The confidence interval spans 25 cM - 34 cM. SWR/Ola-derived alleles at Nhil2 confer increased liver iron content. Tfrc (21 cM) and E330017A01Rik are potential candidate genes for Nhil2.

Highly significant linkage to liveriron in males mapped to 47 cM on mouse Chromosome 7 near D7Mit71 (LOD=4.4). This locus showed suggestive linkage to liver iron in the combined set and is named Nhil3 (non-heme iron levels 3). The confidence interval spans 34 cM - 54 cM. SWR/Ola-derived alleles at Nhil3 confer increased liver iron in males. Hamp1 (11 cM) and Hamp2 (11 cM) are potential candidate genes for Nhil3 and both display increased expression in SWR/Ola mice compared to C57BL/6JOla.

Highly significant linkage to spleen iron mappedto14 cM on mouse Chromosome 8 near D8Mit4 (LOD=5.2). This locus is named Nhil4 (non-heme iron levels 4). The confidence interval spans 6 cM - 23 cM. SWR/Ola-derived alleles at Nhil4 confer increased spleen iron content.

Highly significant linkage to spleen iron mapped to 56 cM on mouse Chromosome 9 near D9Mit182 (LOD=14.3). The confidence interval spans 49 cM - 61 cM. This locus is named Nhil5 (non-heme iron levels 5) and also exhibits significant linkage to spleen iron in the male subset (LOD=10.8). SWR/Ola-derived alleles at Nhil5 confer increased spleen iron content. A potential candidate gene for Nhil5 is Trf at 56 cM. 1300017J02Rik, a downstream transferrin-like gene, is also a potential candidate for Nhil5.

Suggestive linkage to liver iron content in females mapped to 38 cM on mouse Chromosome 8 (D8Mit195, LOD=3.1), 50 cM on mouse Chromosome 11 (D11Mit36, LOD=3.4), and 38 cM on mouse Chromosome 19 (D19Mit37, LOD=3.3).
