Mapping and Phenotype information for this QTL, its variants and associated markersJ:115149Linkage analysis was performed on 289 animals from a (C57BL/6J x DBA/2J)F2 intercross to identify QTLs associated with trabecular bone traits. The proximal tibial metaphyses were phenotyped in 200-day old animals. 97 microsatellite markers at an average spacing of 15 - 20 cM were used for the genome scan. Genomic regions with significant linkage were confirmed using 20 BXD RI strains. A total of 693 animals were analyzed. Significant linkage to bone surface (LOD=2.8) and structure model index (SMI) (LOD=4.4 in males) mapped to 43 cM on mouse Chromosome 1. This locus is named Tbt1 (trabecular bone traits 1). Tbt1 explains 4.2% of the bone surface variance and 13% of SMI variance. C57BL/6J-derived alleles at Tbt1 confer increased bone surface (in both sexes) while DBA/2J-derived alleles increase SMI in males. Previously identified bone QTLs mapping near Tbt1 include Vtbt1 (37 cM), Tlln (50 cM), and Bntq1 (74 cM).Tbt2 (trabecular bone traits 2) mapped to 44 cM - 60 cM on mouse Chromosome 5 and affects bone traits in males. This locus is linked to bone volume fraction (LOD=4.6; explains 20% of variance), bone volume (LOD=4.1; explains 16.6% of variance), connectivity density (LOD=4.8; explains 15.2% of variance), and trabecular number (LOD=4.9; explains 15.8% of variance). DBA/2J-derived alleles at Tbt2 confer increased trabecular bone trait values in male animals. Bmd2 (42 cM) is a previously identified QTL mapping near Tbt2.Significant linkage to trabecular thickness (LOD=5.9; explains 22.4% of variance) and bone surface fraction (LOD=6.9; explains 34.8% of variance) in males mapped to 14 cM - 20 cM on mouse Chromosome 6. This locus is named Tbt3 (trabecular bone traits 3). C57BL/6J-derived alleles at Tbt3 confer increased trabecular thickness while DBA/2J-derived alleles increase bone surface fraction in male animals. Igf1sl1 (51 cM) is a previously identified bone trait QTL near Tbt3. Significant linkage to degree of anisotropy (LOD=3.6; explains 5.6% of variance) mapped to 37 cM on mouse Chromosome 8. This locusis named Tbt4 (trabecular bone traits 4). C57BL/6J-derived alleles at Tbt4 confer increased anisotropy. Tbt4 is also significantly linked to connectivity density (LOD=4.4; explains 11.5% of variance), bone surface (LOD=3.4); explains 9.3% of variance), and trabecular number (LOD=4.4; explains 12.1% of variance) in males. DBA/2J-derived alleles at Tbt4 confer increased connectivity density, bone surface, and trabecular number in males. Bntq9 (57 cM) is a previously identified bone trait QTLnear Tbt4.Suggestive linkage to bone volume (LOD=2.8) and bone volume fraction (LOD=3.2) mapped to 48 - 50 cM on mouse Chromosome 9. Bmd10 (48 cM), Vtbt8 (48 cM), and Fembrs6 (43 cM) are previously identified bone trait QTLs near this locus.Suggestivelinkage to total volume in males (LOD=2.8) and SMI in females (LOD=3) mapped to 38 - 53 cM on mouse Chromosome 12. Vtbt14 (53 cM) and Bntq13 (53 cM) are previously identified bone trait QTLs mapping near this locus. |
