Mutation detailsThe transgene expresses Cre recombinase under control of the promoter region of the gene encoding solute carrier family 5 (sodium/glucose cotransporter), member 2, including 1952 bp of 5'-flanking sequence, the first exon (containing a mutated translation initiation codon), first intron and 23 bp of the second exon. X-gal staining of multiple tissues from mice doubly transgenic for this transgene and for the E. coli beta-galactosidase gene (lacZ) preceded by a loxP-flanked stop sequence (knocked into the Gt(ROSA)26Sor locus) demonstrated beta-galactosidase activity only in the kidney. Immunohistochemical analysis of X-gal-stained kidney cryostat sections for markers specific to different regions of the nephron showed colocalization of X-gal staining only with anti-megalin antibody, which identifies the proximal tubules.
