Mutation details: The transgene is a 6.2 kb human genomic DNA fragment containing the entire lecithin-cholesterol acyltransferase (LCAT) gene plus 851 bp of 5' and 1,134 bp of 3' flanking sequence. Based on Southern blot analysis, hemizygous mice of this founder line harbor 15 transgene copies. The plasma level of LCAT in mice with this transgene is 11-fold that of endogenous LCAT in non-transgenic mice. Northern blot analysis of various tissues from transgenic mice demonstrates expression of LCAT primarily in the liver, with much weaker expression in testis, brain, heart and lung.
