Mapping and Phenotype information for this QTL, its variants and associated markers

Genetic loci associated with body composition were mapped using 369 animals from a (C57BL/6J x CASA/Rk)F2 intercross. A panel of 255 polymorphic markers at an average spacing of 5.9 cM was used for the genome scan. Parental strain C57BL/6J displays decreased activity and increased body weight, lean tissue mass, abdominal fat mass, body length, bone mineral density (BMD), bone area, and bone mineral content (BMC) compared to parental strain CASA/Rk. Males from C57BL/6J display increased body mass index (BMI) and total body fat tissue mass (FTM) compared to males from CASA/Rk.

Linkage total body fat tissue mass mapped to 0 cM (Ftm2, LOD=4.36) on mouse Chromosome 6. Ftm5 mapped to 13 cM (LOD=3.55) for body fat tissue mass in females only. Pbft1 (percent body fat 1) mapped to 5 cM with LOD=5.55. Pbft3 is associated with percent body fat in females and mapped to 17 cM with LOD=5.15. Homozygosity for CASA/Rk-derived alleles at Pbft3 confers increased percent body fat in female mice. Linkage to mesenteric andomental fat pad weight mapped to 1 cM and is named Mofp2 (LOD=4.4). Gonadal fat pad weight mapped to13 cM (Gf1, LOD=4.59) in both sexes and mapped to 17 cM (Gf6, LOD=4.34) in females. Linkage to retroperitoneal fat pad weight mapped to 1 cM (Rfpw2, LOD=5.14) in both sexes and mapped to 16 cM (Rfpw6, LOD=4.48) in females. Linkage to bone mineral content,named Bmc2, mapped to 28 cM with LOD=3.92. Linkage to bone mineral density mapped to 28 cM (Bmd29, LOD=3.79) in both sexes and to 35 cM (Bmd33, LOD=3.64) in females. The leptin gene (Lep) at 10 cM is a potential candidate.
