Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 328 (MRL/MpJ x CAST/EiJ)F2 animals to identify novel genetic loci associated with bone size and density traits. Parental strain CAST/EiJ displays decreased body size, femur length, femur periosteal and endosteal circumference, and mid-diaphyseal femur bone mineral density (BMD) compared to parental strain MRL/MpJ. A panel of 86 polymorphic markers at an average spacing of 17 cM was used for the genome scan.

A novel QTL for femur BMD mapped to 24 cM on mouse Chromosome 10 near D10Mit31 (LOD=3.4). This locus explains 8.9% of the variance and is designated Bmd38 (bone mineral density 38). CAST/EiJ-derived alleles at Bmd38 confer increased bone density. Bmd38 interacts with loci on mouse Chromosomes 5 (45 cM), 8 (15 cM), 14 (10 cM), and 19 (0 cM).

Suggestive linkage to femur BMD mapped to 22.7 cM on mouse Chromosome 15 near D15Mit115. MRL/MpJ-derived alleles at D15Mit115 confer increased bone density. This locus displays significant interaction with mouse Chromosome 19 at25 cM (LOD=4). Pcir3 (33.4 cM) is a previously identified QTL mapping near this region.

Significant linkage to femur periosteal circumference mapped to 31 cM on mouse Chromosome 9 near D9Mit336 (LOD=3.5). This locus explains 7.5% of the variance and is designated Pstc6 (periosteal circumference 6). MRL/MpJ-derived alleles at Pstc6 confer increased femur periosteal circumference. Pstc6 also exhibits significant linkage to femur endosteal circumference (LOD=5.1), forearm total BMD (LOD=4), forearm periosteal circumference (LOD=4.2), and forearm endosteal circumference (LOD=4.2). Pstc6 interacts with loci on chromosomes 2 (75 cM), 3 (20 cM), and Bmd38 on chromosome 10.

Suggestive linkage to femur periosteal circumference mapped to mouse Chromosome 2 near D2Mit411 (LOD=3). MRL/MpJ-derived alleles at D2Mit411 confer increased periosteal circumference. This locus explains 11% of the variance.
