Mapping and Phenotype information for this QTL, its variants and associated markers

J:118316

Linkage analysis was performed using 35 BXD (B=C57BL/6J; D=DBA/2J) recombinant (RI) strains to identify QTLs associated with basolateral amygdala complex (BLAc) volume and cell number. A total of 96 females and 103 males were analyzed at an average age of 96 days. A panel of 3,795 informative SNPs and microsatellite markers were genotyped. Parental strain C57BL/6J and DBA/2J do not display significant differences in BLAc volume or cell number. However, BXD RI strains vary widely in BLAc volume and cell count.

Significant linkage to BLAc volume mapped to mid-chromosome 8 between 65 Mb - 75 Mb (LRS=14). This locus is named Blacv1 (basolateral amygdala complex volume 1). Adjusting for brain weight weakens linkage to a suggestive LRS=10.7 while adjusting for body weight strengthens the linkage to LRS=15. This locus also shows suggestive linkage to cocaine-induced open field behavior. DBA/2J-derived alleles at Blacv1 confer increased BLAc volume in all analyses. Cbm2 at 45 cM is a previously identified brainvolume QTL near Blacv1. Large (34 cM; 75.7 Mb), Fcho1 (74.6 Mb), Sfrs14 (73.1 Mb), Ssbp4 (73.5 Mb), Ncan (73.0 Mb), Spock3 (65.8 Mb), and Zfp617 (74.8 Mb) are potential candidate genes for Blacv1.

Suggestive linkage to BLAc volume mapped to mouse Chromosome 1 between 50 Mb - 60 Mb (LRS=9) and mouse Chromosome 19 between 22 Mb - 28 Mb (LRS=9.2). These loci disappear when brain weight is taken into account and are weakened when adjusted for body weight.

Suggestive linkage to glial cell density in the BLAc mapped to mouse Chromosome 11 with LRS=11.4 between 90 Mb - 105 Mb. This locus approaches significant linkage when adjusted for brain weight (LRS=14.8). Neurod2 (98.1 Mb), Dlg4 (69.8 Mb), Plxdc1 (97.7 Mb), Igf2bp1 (95.7 Mb), and Thra (98.5 Mb) are potential candidate genes mapping near this locus.

Suggestive linkage to neural (LRS=11.4) and glial cell density (LRS=10.2) mapped to mouse Chromosome 13 between 90 Mb - 105 Mb. Hipp3 (31 cM) is a previously identified hippocampal weight QTL mapping nearby.
