Mapping and Phenotype information for this QTL, its variants and associated markersJ:118951Congenic line analysis was used to investigate genetic loci involved in diabetes susceptibility, macrophage STAT5 dysfunction, and GM-CSF production. Diabetes-resistant congenic line NOD.LC11 carries C57L-derived DNA on mouse Chromosome 11 encompassing the Idd4 locus and Stat5a/Stat5b/Stat3 genes on a NOD genetic background. Congenic subline NOD.LC11f carries C57L-derived DNA at Csf2 (29 cM), Idd4a (43 cM), and Idd4b (48 cM) while NOD.LC11b carries C57L-derived DNA only at Csf2. Both lines exclude C57L-derived Stat5a/Stat5b/Stat3 genes. NOD.LC11b exhibits a greater incidence of diabetes (27%) compared to NOD.LC11f (19%) indicating the presence of a diabetes susceptibility locus near Csf. This locus is named Idd4c and also exhibits linkage to decreased GM-CSF production and increased STAT5 phosphorylation in unactivated macrophages. |
