QTL analysis was performed on 334 (C57BL/6J-Apoetm1Unc x C3H/HeJ-Apoetm1Unc)F2 animals to identify QTLs associated with abdominal fat mass. A panel of 1,032 SNPs at an average spacing of 1.5 cM was used for the genome scan. When fed a Western diet for 16 weeks, females from parental strain C3H/HeJ-Apoetm1Unc displays increased adiposity and increased gonadal fat mass compared to females from C57BL/6J-Apoetm1Unc and the opposite is observed in males. Analysis of 23,574 liver transcripts was used to identify eQTLs coincident with the fat mass QTLs.

Significant linkage to gonadal fat mass mapped to 81 cM on mouse Chromosome 3 near rs3697892 (LOD=6.2) and is designated Gofm1 (gonadal fat mass 1). C57BL/6J-derived alleles at Gofm1 confer increased fat mass in females. The cis-eQTL at this locus contains 4 genes. H2afz, Sgms2 (formerly 4933405A16Rik), and Edg7 (70.5 cM) are located near Gofm1 and are considered strong candidate genes.

Gofm2 (gonadal fat mass 2) mapped to 64 cM on mouse Chromosome 5 near rs3662726 (LOD=7.6). C57BL/6J-derived alleles at Gofm2 confer increased fat mass in females whereas C3H/HeJ-derived alleles confer increased fat mass in males. The colocalizing cis-eQTL contains 14 genes.

Gofm3 (gonadal fat mass 3) mapped to 7 cM on chromosome 11 near rs3663826 (LOD=5.1). This locus has a stronger effect in females compared to males with C57BL/6J-derived alleles conferring increased fat mass. There are 7 genes in the colocalizing cis-eQTL. Vrk2 is considered a strong candidate gene for Gofm3. Previously identified abdominal fat weight QTL Afw5 (10 cM) is located near Gofm3.

A female-specific locus for gonadal fat mass, Gofm4, mapped to 45 cM on chromosome 19. C3H/HeJ-derived alleles at Gofm4 confer increased fat mass in females. Thecolocalizing cis-eQTL contains 16 genes. Cyp2c40 and 4833409A17Rik are considered strong candidate genes for Gofm4.

Suggestive linkage to gonadal fat mass mapped to 64 cM on mouse Chromosome 1 near rs3688042 (LOD=5). C3H/HeJ-derived alleles at this locus confer increased fat mass in females. The cis-eQTLs colocalizing here contains 11 genes, of which Arpc5 (64.3 cM) is a strong candidate.
