Linkage analysis was performed on 25 BXD (B=C57BL/6J; D=DBA/2J) recombinant inbred (RI) strains to identify QTLs associated with intersession habituation in mice. Approximately 19-26 male animals from each RI line were tested in a novel environment once a day over a period of 3 days. Males from parental strain DBA/2J display decreased exploratory behavior in a new environment whereas males from parental strain C57BL/6J display increased exploratory behavior.

A single locus on mouse Chromosome 15 between 32 cM and 46 cM is linked to habituation (LOD=8.8). This locus is named Hbtq (habituation QTL) and explains 80% of the genetic variance. Hbtq was confirmed in 72 males from a (C57BL/6J x DBA/2J)F2 intercross with linkage at D15Mit144 (p=0.0036). Homozygosity for C57BL/6J-derived alleles at Hbtq is associated with increased habituation. Eila3 (28 cM), Axtav, Axtofa5, Cocrb14 (39.6 cM), and Cocia2 (47 cM) are previously identified locomotor activity QTLs mapping near Hbtq.
