Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 662 female animals from a (NZB/BlNJ x RF/J)F2 intercross to identify QTLs associated with femur mechanical traits totaling 23 phenotypes. A panel of 89 polymorphic markers at a resolution of 10 cM was used for the genomescan. Female F2 animals were analyzed at 10 weeks of age. Loci reaching the significance threshold are reported below.

Linkage to femur cortical thickness mapped to 95 cM on mouse Chromosome 1 with LOD=3.4. This locus is named Fmtq1 (femur mechanical trait QTL 1).

On mouse Chromosome 3, linkage to femur cortical thickness (LOD=3.9) and volumetric BMD (LOD=4.7) mapped to 10 cM. This locus is named Fmtq2 (femur mechanical trait QTL 2).

Significant linkage to femur ultimate load mapped to 65 cM on mouse Chromosome 4 (LOD=8.5). This locus is named Fmtq3 (femur mechanical traits QTL 3). Fmtq3 is also linked to femur periosteal perimeter (LOD=7.2), cortical area (LOD=6.7), cortical thickness (LOD=7.8), and midshaft cortical BMD (LOD=4.1). NZB/BlNJ-derived alleles at Fmtq3 confer decreased femur ultimate load.

Significant linkage to femur antero-posterior diameter mapped to 15 cM on mouse Chromosome 5 (LOD=6.2). This locus is named Fmtq4 (femur mechanical traits QTL 4). Fmtq5 (femur mechanical traits QTL5) was detected at 30 cM in linkage to femur ultimate load (LOD=3.2) and work to failure (LOD=4.1). Significant linkage to femur latero-medial diameter mapped to 55 cM with LOD=4.0 and is named Fmtq6 (femur mechanical traits QTL 6). Periosteal perimeter exhibited significant linkage to a locus at 80 cM (LOD=4.1) and is designated Fmtq7.

On mouse Chromosome 6, linkage to femur latero-medial diameter mapped to 25 cM with LOD=3.7. This locus is named Fmtq8 (femur mechanical traits QTL 8).

Significant linkage to femur periosteal perimeter mapped to 55 cM on mouse Chromosome 7 (LOD=14). This locus is named Fmtq9 (femur mechanical traits QTL 9). Fmtq9 is also linked to femur work to failure (LOD=3.8), cortical area (LOD=4.2), latero-medial diameter (LOD=4.5), anterior-posterior diameter (LOD=7.4), cortical thickness (LOD=3.6), and volumetric BMD (LOD=4.1).

On mouse Chromosome 8, linkage to femur latero-medial diameter mapped to 0 cM with LOD=4.2. This locus is named Fmtq10 (femur mechanical traits QTL 10).

Femurstrength mapped to 25 cM on mouse Chromosome 9 with LOD=3.3. This locus is named Fmtq11 (femur mechanical traits QTL 11). Fmtq12 exhibits maximum linkage to femur cortical thickness (LOD=9.3) at 45 cM. Fmtq12 is also linked to femur ultimate load (LOD=7.6), work to failure (LOD=5), cortical thickness (LOD=5.7), and antero-posterior diameter (LOD=4.5).

Linkage to femur latero-medial diameter (LOD=6.5) and volumetric BMD (lod=3.2) mapped to 65 cM on mouse Chromosome 10. This locus is named Fmtq13 (femurmechanical traits QTL 13).

Linkage to cortical thickness mapped to 20 cM on mouse Chromosome 11 (LOD=5.9). This locus is named Fmtq14 (femur mechanical traits 14). Linkage to femur stiffness (LOD=3.3), volumetric BMD (LOD=6.2), and midshaft cortical BMD(LOD=3.3) mapped to 10 cM and is named Fmtq15. Linkage to femur periosteal perimeter (LOD=26) mapped to 65 cM on mouse Chromosome 11 explaining 8.9% of the variance and is designated Fmtq16 (femur mechanical traits QTL 16). This locus also exhibits significant linkage to femur latero-medial diameter (LOD=13.1), antero-posterior diameter (LOD=8.8), femur ultimate load (LOD=4), and cortical area (LOD=8.4).

Significant linkage to femur periosteal perimeter (LOD=7.5) and latero-medial diameter (LOD=4.1) mapped to55 cM on mouse Chromosome 12. This locus is named Fmtq17 (femur mechanical traits QTL 17).

Fmtq18 (femur mechanical trait QTL 18) mapped to 70 cM on mouse Chromosome 13. This locus is associated with femur cortical thickness (LOD=5.9).

Fmtq19 (femur mechanical trait QTL 19) mapped to 25 cM on mouse Chromosome 15. This locus is associated with femur ultimate load (LOD=6.7), cortical area (LOD=3.3), and latero-medial diameter (LOD=3.2). A more distal locus named Fmtq20 mapped to 50 cM with significant linkage to femur periosteal perimeter (LOD=8.2).

On mouse Chromosome 18, linkage to femur cortical thickness (LOD=4.8), volumetric BMD (LOD=4.9), and midshaft cortical BMD (LOD=5.7) mapped to 55 cM. This locus is named Fmtq21 (femur mechanical trait QTL 21).
