Mapping and Phenotype information for this QTL, its variants and associated markersJ:122126Linkage analysis was performed on 152 female (B6.KK-Apoa2b x RR)F2 animals to identify modifiers of Apoa2 on mouse Chromosome 1 with respect to plasma total cholesterol concentration and body weight. Females from parental strain RR/Sek have increased body weight compared to females from parental strain B6.KK-Apoa2b. Animals were phenotyped at approximately 124 days of age. A suggestive locus for plasma total cholesterol mapped between D2Mit274 (52.5 cM) and D2Mit285 (86 cM) on mouse Chromosome 2 with LOD=2.8. This locus explains 9.8% of the variance and RR-derived alleles at D2Mit285 confer increased plasma total cholesterol.A second suggestive locus for plasma total cholesterol mapped near D3Mit25 (29.5 cM) on mouse Chromosome 3 with LOD=3.5. This locus explains 11.3% of the variance with C57BL/6JJcl-derived alleles at D3Mit25 conferring increased plasma total cholesterol. Cq3 is a previously identified cholesterol QTL at 49.7 cM. However, the 95% confidence intervals of Cq3 and the D3Mit25 locusdo not overlap. Hdlq21 (56 cM) and Tchol1 (49.2 cM) are also previously identified cholesterol QTLs in the general vicinity.Significant linkage to body weight mapped to 50 cM on mouse Chromosome 3 near D3Mit102 (LOD=5.2). This locus explains 14.4% of the F2 body weight variance and is named Bwq8 (body weight QTL 8). RR/Sek-derived alleles at Bwq8 confer decreased body weight. The QTL spans 38 cM to 63 cM and overlaps with Cq3 and the D3Mit25 locus. Afw1 (30 cM), Afpq1 (26 cM), Wt10q2 (61 cM), Batq2 (55cM), and Iba2 (49.7 cM) are previously identified body weight QTLs mapping to chromosome 3. Potential candidate genes for Bwq8 include Fabp2 (55 cM) and Kcna3 (52.3 cM). |
