Chromosome substitution strains (CCSs) derived from A/J donor chromosomes on a C57BL/6J genetic background were used to map QTLs for conditioned fear response. Animals aged 48 - 80 days were assessed for freezing to context and freezing to tone before foot shock. A total of 485 CCS mice were phenotyped, along with 82 C57BL/6J and 17 A/J. CCS strains for chromosomes 2 and 8 were not available for study. Recipient strain A/J displays increased freezing to context and altered context, as well as increased pre-training freezing compared to recipient strain C57BL/6J. A/J females displayed significantly increased pre-training freezing compared to A/J males.

CCS-7 and CCS-14 displayed significantly lower freezing to context compared to C57BL/6J, indicating A/J-derived QTL(s) on mouse Chromosomes 7 and 14 that decrease this trait relative to C57BL/6J. These loci are named Frct1 (fear conditioning traits 1) and Frct2 (fear conditioning traits 2), respectively.

Significantly higher freezing to tone was observedinCCS-10 and CCS-18 indicating A/J-derived QTL(s) on mouse Chromosomes 10 and 18 that increase this trait relative to C57BL/6J. These loci are named Frct3 and Frct4, respectively. Frct3 accounts for 24% of the genetic variance in freezing to tone. A previously identified QTL named Exq1 mapped to 67.5 cM on chromosome 10 by Zhang et al (2005) in A/JOlaHsd and C57BL/6JOlaHsd mice. Significantly lower freezing to tone was observed in CCS-12. This locus is named Frct5

CCS-3 displayed significantly higher pre-training freezing and suggestive linkage to decreased freezing to tone and context. CCS-1 also displayed significantly higher pre-training freezing. Authors indicate this is a measure of baseline locomotor activity and not related to fear learning.
