Mutation details: The transgene contains a human APP minigene encoding the Indiana mutation (V717F) which is associated with familial Alzheimer disease. The construct contained APP introns 6-8, allowing alternative splicing of exons 7 and 8. The human PDGF beta-chain promoter drives expression, and the construct also contains the SV40 polyadenylation signal. Amyloid-beta ELISA on whole brain homogenates indicates higher cerebral beta-amyloid production than in Tg(APPV717F)109Ili mice expressing the same construct.
