Linkage analysis was performed on 24 BXD (B=C57BL/6J, D=DBA/2J) recombinant inbred (RI) strains to identify genetic loci involved in stress and despair as measured by duration of immobility during the tail suspension test. Two separate tail suspension trials were performed and each trial lasted 5 minutes. Parental strain C57BL/6J displays greater duration of immobility and lesser frequency of immobility compared to parental strain DBA/2J. Investigators observed different genetic contributions in trial 1 and trial 2 and maximum linkage was observed using a 20% mobility threshold.

On mouse Chromosome 4, a locus associated with trial 1 duration of immobility showed linkage to SNP marker rs13477796 (78.5 Mb, LOD=4.56). This locus spans approximately 20 Mb and is named Tsti1 (tail suspension test immobility 1). Potential candidate genes found in this interval include Ptprd (38 cM), Tyrp1 (38 cM), and Mpdz (38.6 cM). Interestingly, peak linkage for Tsti1 occurs within an intron of Mpdz and this gene is knownto carry several non-conservative polymorphisms between C57BL/6J and DBA/2J. In addition, Ptprd shows positive correlation with the tail suspension test phenotype. A distal locus associated with trial 1 mobility frequency mapped near SNP marker rs6202860(LOD=2.55).

Duration of immobility in trial 1 also mapped to mouse Chromosome 15 near SNP marker rs13459176 (3.2 Mb, LOD=3.41) within a Sepp1 (5.9 cM) exon. This locus spans <13 Mb and is named Tsti2 (tail suspension test immobility 2). Potential candidate genes found in this interval include Prkaa1, Gdnf, Slc1a3 (6.7 cM), and Pdzd2. The gene expression pattern of Slc1a2, a gene on chromosome 2 that interacts with Slc1a3, displays positive correlation with the tail suspension test phenotype in trial 2.

Linkage to duration of immobility in trial 2 mapped to mouse Chromosome 11 near SNP marker rs13481087 (LOD=3.59) within an Atp1b2 (40 cM) intron. This locus is named Tsti3 (tail suspension test immobility 3).

Linkage to duration of immobility during trial 2 mapped to mouse Chromosome 18 near SNP marker gnf18.027.000 (LOD=2.19). This locus also exhibits linkage to frequency of mobility during trial 1 and is named Tsti4 (tail suspension test immobility 4).
