Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 180 animals from 34 BXD (B=C57BL/6J, D=DBA/2J) recombinant inbred (RI) strains to identify QTLs associated with thalamus and cortex gray matter volume. Parental strain C57BL/6J displays significantly increased thalamus volume and cortex gray matter volume compared to DBA/2J. Experimental animals ranged between age 31 days and 493 days. A panel of 781 polymorphic markers was used for the genome scan.

A locus on mouse Chromosome 2 near D2Mit42 (47 cM) shows suggestive linkage to cortex gray matter volume with LOD=2.32. The QTL interval is flanked by D2Mit340 (68.9 cM) and D2Mit493 (72 cM). Flrt3 and 2900060K15Rik map near this interval. The gene expression pattern exhibits correlation to the cortex volume phenotype.

Suggestive linkage to cortex gray matter volume mapped to mouse Chromosome 8 near D8Mit312 (45 cM, LOD=2.8).A potential candidate gene mapping near this interval is Ranbp10 (formerly 4432417N03Rik). The expression pattern of this gene shows correlation to the cortex volume phenotype.

Suggestive linkage to thalamus volume mapped to mouse Chromosome 10 near D10Mit14 (LOD=2.22). A potential candidate gene mapping near this locus is Casc5 (formerly 2310043D08Rik-*Note: mapping records in MGD place Casc5 on chromosome 2). The expression pattern of this gene is correlated to the thalamus volume phenotype.

Suggestive linkage to thalamus volume mapped to mouse Chromosome 11 near D11Mit19 (13 cM, LOD=2.86). Vamp2 and Ccdc104 (formerly 4931428D14Rik) are potentialcandidate genes mapping near this locus. The expression pattern of these genes is correlated to the thalamus volume phenotype.

Suggestive linkage to thalamus volume (LOD=2.63) and cortex gray matter volume (LOD=2.33) mapped to a common locus on mouse Chromosome 16 near D16Mit100 (8.5 cM). The QTL interval is flanked by D16Mit9 (4 cM) and D16Mit167 (26.5 cM). This locus also shows linkage to a behavior phenotype, specifically Morris water maze search time preference. This locus is designated Bbq1 (brain and behavior QTL 1). Fgd4 and Tnfrsf17 are potential candidate genes for Bbq1. These genes map within the QTL interval and the brain gene expression patterns show correlation to thalamus and cortex gray matter volume phenotypes.

Suggestive linkage to cortex gray matter volume mapped to mouse Chromosome 19 near D19Mit40 (25 cM, LOD=3.13). The QTL interval is flanked by D19Mit16 (15 cM) and D19Mit63 (24 cM). Akt1 and C130032J12Rik were proposed as possible candidate genes. The gene expression patterns correlate to the cortex gray matter phenotype. (*Note: Mapping records in MGD place Akt1 on mouse Chromosome 12 and C130032J12Rik on mouse Chromosome 14).
