Mapping and Phenotype information for this QTL, its variants and associated markersJ:125685Linkage analysis was performed on more than 200 animals from a (BALB/c-Irf2tm1Mak x C57BL/6-Irf2tm1Mak)F2 x C57BL/6-Irf2tm1Mak backcross to identify genetic modifiers of psoriasis-like skin disease. Parental strain C57BL/6-Irf2tm1Mak displays accelerated psoriasis-like skin disease due to increased Ifna/Ifnb responses whereas parental strain BALB/c-Irf2tm1Mak does not develop skin disease. Genome scan was performed using polymorphic markers spaced approximately 20 cM - 30 cM apart. Significant linkage to skin disease severity mapped to 42.5 cM on mouse Chromosome 4 near D4Mit27 (LOD=8.5). This locus also shows weak linkage to defective CD4+ dendritic cell development and is named Psds1 (psoriasis-like skin disease severity 1). The Psds1 QTL interval is flanked by markers D4Mit327 (42.5 cM) and D4Mit187 (49.6 cM). Potential candidate genes found in this interval include the Ifna/Ifnb gene cluster (42.6 cM) as well as Jun (44.6 cM). An epistatic interacting locus was detected at 38 cM on mouse Chromosome 16 near D16Mit5. This locus is named Psds3 (psoriasis-like skin disease severity 3). Animals homozygous for the Irf2tm1Mak mutation, homozygous for C57BL/6 alleles at Psds1, and heterozygous for C57BL/6 and BALB/c alleles at Psds3 displayed the severest skin disease. Significant linkage to defective CD4+ dendritic cell development mapped to an interval between D4Mit204 (61.9 cM) and D4Mit54 (66 cM) with LOD=4.4. This locus is named Psds2 (psoriasis-like skin disease severity 2). Significantlinkage to skin disease severity also mapped to mouse Chromosome 10 with LOD=6.4 between D10Mit35 (69 cM) and D10Mit297 (70 cM). This locus is named Psds4 (psoriasis-like skin disease severity 4). A potential candidate gene for Psds4 is Stat2 at 70 cM. |
